Cyclodextrin Glycosyltransferase Scholarly Peer-review Journal
Cyclodextrin glycosyl transferase (also Cyclodextrin glucosyltransferase or Cyclodextrin glucanotransferase) or CGTase for brief (EC 2.4.1.19) may be a bacterial enzyme belonging to an equivalent family of the α-amylase specifically referred to as glycosyl-hydrolase family 13. This peculiar enzyme is capable of catalyzing quite one reaction with the foremost important being the synthesis of non-reducing cyclic dextrins referred to as cyclodextrins ranging from starch, amylose, and other polysaccharides. CGTase is an enzyme common to several bacterial species, especially of the Bacillus genus (e.g. B. circulans, B. macerans and B. stearothermophilus), as well as to some archaea, but it is not known to be present in any other species.
In contrast, thermolabile CGTases are isolated with very interesting specificities, just like the production of one size of cyclodextrin. To take advantage of such a specificity in an industrial context, thermostability will need to be engineered into these thermolabile CGTases through site-directed mutagenesis. This is a challenging task, which requires foremost detailed insight into the enzyme’s individual structure and function.
