Plasmid DNA Innovations

Plasmids have a wide range of functions. The genes carried in plasmids provide bacteria with genetic advantages, such as antibiotic resistance. They may contain genes that improve the endurance of an organism, either by defending the host cell by producing toxins or by killing other organisms. Few plasmids facilitate the process of replication in bacteria. The demand for plasmid DNA (pDNA) has vastly inconceivably expanded over the past decade in response to noteworthy advances that have been made in its application for gene therapy and vaccine development. Plasmid DNA-based vaccines are experiencing resurgence due to achievement with prime-boost immunization strategies. The challenge has always been poor efficiency and release of pDNA. Plasmid DNA-based vaccines have conventionally required milligram scale of GMP-grade product for immunization due to the relatively low duration and efficacy of gene expression. However, efforts to increase pDNA vaccine efficiency are evolving in genetic manipulations of bacterial host, enhancements in product recovery and novel delivery methods.
