Mutagenesis Innovations
The site-directed mutagenesis is an artificial (in vitro) technique for introducing mutation or alteration into the target (known) DNA sequence by using artificial techniques such as PCR a mutation or alteration can be inserted into the target-DNA sequence. In vitro mutagenesis is used to persistently to change genetic information. Analysis of the subsequent changes in gene expression and gene products helps elucidate the functional effect of the mutation. Plant mutagenesis in crop improvement, the high selection pressure applied in rice breeding since its domestication thousands of years ago has caused a narrowing in its genetic variability. Successful gene modifications have been obtained by unsystematic and targeted mutations. Furthermore, following mutation initiation pathways, techniques have been applied to identify mutations and the molecular control of DNA damage repair mechanisms in the rice genome. This review highlights findings in generating rice genome resources showing strategies applied for variability increasing, detection and genetic mechanisms of DNA damage repair.
