Future Of Glycomics

Membrane proteins mediate cell–cell interactions and adhesion, the transfer of ions and metabolites, and therefore the transmission of signals from the extracellular environment to the cell interior. The extracellular domains of most cell wall proteins are glycosylated, often at multiple sites. there's a growing awareness that glycosylation impacts the structure, interaction, and performance of membrane proteins. the appliance of glycoproteomics and glycomics methods to membrane proteins has great potential. However, challenges also arise from the unique physical properties of membrane proteins. Successful analytical workflows must be developed and disseminated to advance functional glycoproteomics and glycomics studies of membrane proteins. This review explores the opportunities and challenges associated with glycomic and glycoproteomic analysis of membrane proteins, including discussion of sample preparation, enrichment, and MS/MS analyses, with attention on recent successful workflows for analysis of N ‐ and O ‐linked glycosylation of mammalian membrane proteins.
 

Glycosylation is one among the foremost important post-translational modifications of proteins, known to be involved in pathogen recognition, innate immune reaction and protection of epithelial membranes. However, in comparison to the tools and databases available for the processing of high-throughput proteomic data, the glycomic domain is severely lacking. While tools to help the analysis of mass spectrometry (MS) and HPLC are continuously improving, there are few resources available to support liquid chromatography (LC)–MS/MS techniques for glycan structure profiling. Here, we present a platform for presenting oligosaccharide structures and fragment data characterized by LC–MS/MS strategies. The database is annotated with high-quality datasets and is meant to increase and reinforce those standards and ontologies developed by existing glycomics databases.
