Medicinal plants, vegetables and fruits are the sources of giant number of bioactive lead/scaffolds with therapeutic and nutraceutical importance. Bioautography may be a means of target-directed isolation of active molecules on chromatogram. Organic solvents employed in chromatographic separation process are often completely removed before biological detection because these solvents cause inactivation of enzymes and/or death of living organisms. they provide a rapid and straightforward identification of bioactive lead/scaffolds in complex matrices of plant extracts. Bioautography may be a technique to isolate hit(s)/lead(s) by employing an appropriate chromatographic process followed by a biological detection system. This review critically describes the methodologies to spot antimicrobial, antioxidant, enzyme inhibitor lead/scaffolds by employing bioautography. a big number of examples are incorporated to authenticate the methodologies.


In order to see the purity of a- and b-cyclodextrins with reference to one another partition chromatog. on cellulose columns was used (elution with 42:29:29 BuOH-EtOH-H2O, 0.3 atm. atmospheric pressure on the solvent reservoir; 75-90 ml./hr. flow rate). Amts. down to 2.5% a-cyclodextrin (I) in b-cyclodextrin (II) and the other way around might be detected within the eluate fractions with an automatic polarimeter. Amts. down to 0.5% II in I and to 0.3% I in II were traced by concg. the eluant, applying the soln. to paper, and spraying the paper with alc. iodine soln
